Description

Collagens (types I, II, III, IV and V) are synthesized as precursor molecules called procollagens. These contain additional peptide sequences, usually called "propeptides", at both the amino-terminal and the carboxy-terminal ends. The function of these propeptides is to facilitate the winding of procollagen molecules into a triple-helical conformation within the endoplasmic reticulum. The propeptides are cleaved off from the collagen triple helix molecule during its secretion, after which, the triple helix collagens polymerize into extracellular collagen fibrils. These Clones recognize procollagen type I C-peptide. They are suitable for the detection of non-denatured antigen released in tissue culture media and for monitoring of collagen synthesis.

The stock solution (2.0 mg/ml) with Reconstitution Solution should be stored in aliquots at -20°C for 1 year, or should be stored at 4°C for 6 months. Avoid repeated freeze-thaw cycles. The diluted antibody should not be stored.

Reconstitution

Dissolve the lyophilized antibody in 50 µl of Reconstitution Solution. (final concentration: 2.0 mg/ml)

Specificity

  • These products specifically react with procollagen type I C-peptide.
  • These products react with human, bovine, horse and dog antigen.
  • These products do not react with rat and rabbit antigen.

Application

  • These products can be used effectively in studies on the synthetic capacity of collagen by measuring procollagen type I C-peptide in cell culture supernatants.
  • These products can be used for histology on paraffin embedded and frozen tissue sections.
  • These products can be used for Western blotting analysis under non-reducing and non-heating condition.
