Description
This DNA is a vector which is available to enforcement cloning system. This DNA contains chloramphenicol resistant gene and lethal gene rpsL (streptomycin-sensitive ribosomal protein) at downstream region of tryptophan promoter. Since this is designed for multi-cloning site possessing amber mutant and 39 restriction sites on rpsL gene, normal rpsL will not be expressed if a target gene is inserted into these restriction sites. Only targeted clones can be grown, because uninserted clones can not be grown in trpR -, rpsL - and supE - host like E. coli TH2. Therefore, it is easy to select targeted clones.

The restriction endonucleases in parentheses can cleave several sites of pKF3 DNA.
Since the marked (*) restriction endonucleases are affected by dam methylase, those can not cleave dam+ DNA. Takara pKF3 DNA (Cat.# 3100) is methylated by dam methylase.
