Description

pPTR II is an E. coli-Aspergillus shuttle vector which can replicates autonomously in Aspergillus cells. This vector has an ampicillin resistant gene, AmpR, as the selection marker for E. coli and a Pyrithiamine-resistant gene, ptrA, as that for Aspergillus. Since it has cloning sites, containing Hind III, Sma I, and Kpn I on lacZ region, any DNA insertion into this vector can be easily verified using plates containing IPTG and X-Gal.

Intended usage

This is a vector for transformation of Aspergillus species using Pyrithiamine which is metabolic competitive analog of thiamine. It is maintained as a plasmid in Aspergillus cell.

Note

As the presence of thiamine causes the remarkable decrement of selection efficiency by Pyrithiamine, the media and the reagents without thiamine should be used for this transformation system.
