Description
FastPure™ DNA Kit is a spin column type which quickly and easily extracts highly pure genomic DNA from cultured cells or mammalian tissue. The kit makes it possible to obtain high quality genomic DNA by utilizing a newly developed polymer membrane.
First, dissolve the mammalian tissue or cells using the Tissue Lysis Buffer and Proteinase K to prepare a lysate to use with this kit. Using the cartridge with the polymer membrane, collect genomic DNA by following a protocol using a microcentrifuge, making both organic solvent or ethanol precipitation unnecessary . The entire extraction process of the lysate to genomic DNA will take about 30 minutes. The polymer membrane used in this kit is extremely thin, allowing it to have a higher nuclear adsorption compared to the glass fibers commonly used in genomic DNA preparations.
The resulting genomic DNA is high quality and is suitable to use in applications such as PCR, restriction enzyme treatment, or Southern blotting.

Note
- Perform pre-experiment which starting extraction process from tissue amount of 5 mg if the sample is extracted the first time using this kit.
- The cartridges have a limited capacity. When the cartridge is overloaded then the recovery of DNA will be decreased or the worst case would be clogging the cartridge.
- The amount of tissue processed depends on itfs size, type and process condition.
- Tissue Lysis Buffer (TB) might precipitate during storage. To resuspend, heat at 55°C until dissolved. Cool down solution to room temperature before use.
- Lysis Buffer (LB) might precipitate during storage. In such case, dissolve at 37°C, cool down to room temperature before use.
- Avoid using or store LB in highly humid condition.
- Do not mix solution or flow through which contains LB with breach.
- All operation should be performed at room temperature (15 - 30°C). If kit was used in low or high temperature condition, it might not perform as it should be.
- Close the cartridge lid tight for centrifuge.
- Push cartridge deep when setting onto the waste tube.
- Avoid consecutive drive of centrifuge to prevent heat up inside of the centrifuge. Extraction performance might receive negative influence.
- Use centrifuge under the specific condition (cycles, time, etc).
