Description

Dr. GenTLE® (from Yeast) High Recovery is designed to efficiently extract and purify genomic DNA from yeast cells by using both digestion enzyme of cell wall and salting out for DNA purification. Procedure for genomic DNA purification using this kit is as the following; recovery of yeast cells by centrifugation, digestion of yeast cell wall by GenTLE Yeast Solution A (contains cell wall lytic enzyme), cell lysis by GenTLE Yeast Solution B, salting-out for protein removal by adding GenTLE Yeast Solution C and centrifuging, and DNA recovery by isopropanol precipitation.
This kit is useful to isolate genomic DNA, which is available for general genetic engineering, such as Southern blotting, PCR, or restriction enzyme treatment, from various species of yeast listed on the Table A and B in below. For standard experiment, 4 - 10 µg of genomic DNA (35 - 200 kb) can be obtained from 1 - 2×108 yeast cells.

Components

1. GenTLE Yeast Solution A*1 15 ml
2. GenTLE Yeast Solution B *2 3 ml
3. GenTLE Yeast Solution C 6 ml
4. TE Buffer6 ml

*1 GenTLE Yeast Solution A contains enzyme. Avoid mixing vigorously or doing operations which may inactivate enzyme.
*2 If precipitates appeared in GenTLE Yeast Solution B, use after dissolving completely the precipitates by heating up at 37°C.

Storage

4°C
Only GenTLE Yeast Solution B can be stored at room temperature. When stored at low temperature, precipitates may appear. If precipitates appear, dissolve precipitates completely by heating at 37°C before use.
