Protocol of High-Salt Solution for Precipitation (Plant)



  1. Add 0.5 volume of this solution to RNA sample*1, and mix.
  2. Add same volume of isopropanol as the solution added at 1), and mix.
  3. Stand for 10 minutes at room temperature.
  4. Centrifuge at 12,000 rpm for 10 minutes at 4°C.
  5. Discard the supernatant.
  6. Add 1 ml of cold 75% ethanol to the precipitate.
  7. Re-centrifuge at 12,000 rpm for 5 minutes at 4°C.
  8. Discard the supernatant, and dry the precipitate briefly*2.
  9. Dissolve the precipitate in appropriate amount of RNase-free water or TE buffer.
*1 This product is also available for the extraction solution before isopropanol precipitation during RNA extraction procedure by AGPC method.
*2 Do not dry completely the precipitate. It may cause difficulty with dissolving RNA.
