薬剤耐性遺伝子 (Antibiotic Resistance Genes, ARGs)の検出

抗菌薬の発見と使用は20世紀の医学において重要なブレークスルーであり、ヒト臨床分野では罹患率や死亡率を劇的に減らした。農業分野においても抗菌薬は広く利用されており、家畜の成長促進、疾病予防のために抗菌薬が投与されることで、食糧生産量の増加につながっている。抗菌薬には、細菌の細胞壁を傷つけるもの、DNA・RNAあるいはタンパク質合成を阻害するもの、または細菌の代謝や成長を阻害するものがあり、それらの作用機序に基づき複数のクラスに分類されている。いくつかの抗菌薬は狭いスペクトルで特定の細菌種に特異的に作用する一方で、広いスペクトルを持つ抗菌薬は多くの細菌に効果を発揮する。

耐性細菌

抗菌薬に関する新たな問題として耐性細菌の出現がある。細菌は、抗菌薬による選択的な圧力に曝されることで、時間とともに薬剤耐性遺伝子(ARG)を獲得することがある。つまり過度の抗菌薬の使用により、特定の細菌で自発的なARGが出現・獲得する可能性がある。これらの耐性菌が増殖・拡散してしまうと、新たに抗菌薬を開発して対抗する必要がある。現在の薬剤耐性菌に対する対策は、抗菌薬の過度の使用を防ぐことで、細菌が抗菌薬に曝露されるのを制限し、選択圧を最小限にすることである。臨床分野の場合、投薬量が不適切なために起こる過剰使用、大量処方や間違った処方、あるいは不適切な廃棄の結果として細菌が抗菌薬に余計に暴露される。農業分野における抗菌薬の使用もまた細菌がARGを獲得することに寄与している。抗菌薬の過剰使用による家畜体内での耐性菌の出現だけでなく、堆肥が土壌、水、農作物を介して環境中に耐性菌を伝達する運び屋(ベクター)として機能することで、耐性細菌は広範囲に拡散する。そこで近年の取り組みとして、薬剤耐性遺伝子(ARG)を同定し、様々な環境サンプル中のARGを判別し、追跡する様な研究がフォーカスされている。

現在、数百種類の抗菌薬に対して耐性をもたらす約400種類近くのARGが特定されている。そのため、ヒトおよび環境サンプル中のARGを調査するためには、タイプの異なる多数のサンプル中に存在する多様な標的ARG配列を識別するために、ハイスループットリアルタイムPCRスクリーニングを行う必要がある。さらに細菌は常に新しいARGを生み出すため、測定対象となるARGを追加・変更できることが不可欠で、ARGの研究には、ハイスループット性だけでなく柔軟性も兼ね備えたリアルタイムPCRシステムが必要となる。

SmartChip Real-Time PCR Systemは、さまざまなサンプル中のARGをプロファイリングし、追跡するために数多くの研究で使用されている。初めは296個のARGをターゲットとしたパネルを用いて実施され、1チップ当たり最大16個の土壌サンプルをスクリーニング可能な反応条件であった。その後パネルは384個のARGをターゲットとして含むようになり、土壌、水、土砂、肥料、レタス、魚、およびスラッジといった様々なサンプルを対象としている。また、SmartChipシステムの柔軟性とスループット性は、薬剤耐性獲得のメカニズムの研究にも役立てられている。以下にSmartChipシステムによる薬剤耐性遺伝子(ARG)研究の文献例を紹介する。
