製品説明

Primary Cancer Culture Systemは、ヒト腫瘍組織サンプルから初代がん細胞を単離・培養するために標準化されたシステムである。本製品はPrimary Cancer Cell Medium D-ACF、SupplementMixおよびコーティング剤NCCD-Reagentから構成されており、動物由来成分や血清を含んでいない。 元の腫瘍の特性と不均一性を維持した状態でがん細胞の長期培養が可能である。 長期培養を行うことで、悪性細胞を機能的に選択し、初代がん細胞が濃縮された細胞集団を得ることができる。

内容と保存

Primary Cancer Cell Medium D-ACF250 ml：4℃で暗所保存
Primary Cancer Cell Medium D-ACF SupplementMix2 ml：－20℃保存
NCCD-Reagent2 ml：－20℃保存＊

＊ 単品購入も可能。PBS w/o Ca2＋／Mg2＋（製品コード C-40232）にて20倍希釈し、
100 μl／cm2にて培養プレートの表面をコートする。

※ Primary Cancer Cell Medium D-ACFにSupplementMixを混合した後は、4℃で8週間保存可能。

本製品以外に必要な試薬や器具

  • 新鮮な腫瘍組織（0.2～3 g；1 g以上が望ましい）
  • Hanks Balanced Salt Solution (HBSS) with Ca2＋／Mg2＋ w/o Phenol Red
  • Gentamicin (50 mg/ml stock)
  • Phosphate buffered saline (PBS) w/o Ca2＋／Mg2＋（製品コード C-40232）
  • Accumax（Sigma #A7098など）※組織の分散に使用
  • Accutase（製品コードC-41310）※樹立した細胞の継代時に使用
  • メス／ピンセット／はさみ
  • セルストレイナー（ポアサイズ400／100／40 μmなど）
  • ロータリーミキサー
  • 組織培養用フラスコ、デイッシュ
